FIELD: biotechnology.
SUBSTANCE: disclosed is a method for assessing the morphological structure of biofilms of microorganisms, including the formation of a biofilm of microorganisms on a cover glass, for which 50–200 mcl of suspension of mother culture of bacteria, diluted to concentration of 0.5–3.0 by McFarland, applied on a cover glass, incubated for 1 h at 37±2 °C, then the glasses are washed with phosphate buffered saline, immersed with a narrow face in a dense nutrient medium at angle of 40–50° to surface, 10 ml of a liquid nutrient medium is poured into a dish, incubated for 7 days at 37±2 °C with complete replacement of liquid nutrient medium every 24 hours; before staining, the preparation is fixed for 3 minutes in 96% ethanol with subsequent removal of alcohol and gluing with a transparent varnish on a slide, stained with a dye for luminescent microscopy. Structure of the biofilm is visualized using an optical microscope with a luminescent module at magnification x10-100 in an immersion system, and a morphometric study is carried out using the ImageJ ver. 1.52a program.
EFFECT: invention provides evaluation of morphology of biofilm of microorganisms at different stages of its formation, creation of three-dimensional models with saving of research results in the form of graphic files.
