FIELD: biotechnology.
SUBSTANCE: evaluation method of activity of nucleotide excision repair system in mammals is proposed, comprising obtaining a DNA model and its incubation with extract protein of mammal in the presence of matrix oligonucleotide comprising insertion segment based on amidophosphite and also oligo dG tract at 5', followed by detection of the radioactive products obtained as a result of polymerase reaction, with length of 25-36 nucleotides with use of sequencing electrophoresis gel under denaturing conditions, while the evaluation of activity of nucleotide excision repair system is carried out based on the relative intensity of bands corresponding to the radioactive products with length of 25-36 nucleotides.
EFFECT: due to high sensitivity, the method may be used in medicine to evaluate the activity of damage repair system caused by exposure to physical and chemical environmental factors, chemotherapeutic preparations, in extracts of normal and tumor cells.
