SUBSTANCE: method of purification of vaccinia virus, including recombinant its recombinant variants, comprises sonication of the original virus-containing liquid. The resulting suspension is centrifuged at 24,000 rev/min. The precipitate is resuspended in the buffer. It is re-sonicated, treated with modified trypsin at a concentration of 50-75 mcg/ml for 10-30 minutes at a temperature of 37°C. Then the suspension is frozen at -20°C. After thawing, the virus-containing suspension is applied to the 36% sucrose solution and centrifuged at 30,000 rev/min, followed by resuspension of the resulting precipitate in the buffer solution. At that for resuspension of the precipitations containing viral particles, 0.05 M Tris-HCl, buffer with pH 9.0 are preferably used. The modified trypsin is used as salt-free trypsin treated with L-(tosylamido-2-phenyl)-ethyl-chloromethyl ketone.

EFFECT: invention enables to speed up and reduce the cost of the process of obtaining of highly purified vaccinia virus, including its recombinant versions.
