SUBSTANCE: invention can be applied for vitality determination for non-cultivable forms (NF) of Vibrio cholerae O1/O139 in water objects during monitoring surveys, in NF research in tests for non-cultivated state induction, in studies of NF resistance to physical and chemical effects, in other research where vitality confirmation is required with levelling of lack of NF growth in dense nutrient media. Method involves preliminary preparation of reference vegetative cultures of V. cholerae O1 and V. cholerae O139 and test samples, determination of protein in all samples by Lowry protein assay, with parallel production of "crushed drop" preparations out of all samples with acridine orange, registering fluorescence character in luminescent microscope (green fluorescence stands for live culture, red/orange fluorescence corresponds to dead culture), with further determination of ribulose biphosphate carboxylaze (RBPC) activity by radio isotope method with marked bicarbonate. Results are registered in scintillation counter and expressed in pulse.min/mcg of protein. RBPC activity available in a test sample indicates vitality of NF of V. cholerae O1/O139.
EFFECT: vitality determination for the said sample of various ages in aqueous medium of different composition.
