FIELD: chemistry.

SUBSTANCE: invention refers to biotechnology. A method for producing α-ketoglutaric acid provides the aerobic culture of the yeast strain Yarrowia lipolytica VKM Y-2412. The medium has the following composition (g/l): rape oil - 20; (NH4)2SO4 - 6, MgSO4"Ч"7H2O - 1.4, Ca(NO3)2 - 0,8, NaCl - 0.5, KH2PO4 - 2.0, K2HPO4 - 0,2, microelements (mg/l): KJ - 0.1, B3+ - 0.01, Mn2+ - 0.01, Zn2+ - 0.01, Cu2+ - 0.01, Mo2+ - 0.01, Fe2+ - 1.2 mg/l, thiamine - 1.2 mcg/l, distilled water - up to 1 l. That is combined with adding ammonium sulphate additionally in a dose of 1.2 g/l every 20-30 h of the culture process. The first stage of the biomass growth is performed for 44-48 h at pH 4.1-5.0 with aeration at 20-25% of the saturation level. At the second culture stage, in the period from 44-48 h to 70-72 h pH is supposed to maintain at 4.1-5.0. At the third culture stage, in the period from 70-72 h to 188-192 h pH is reduced to 3.0-4.0. Additionally at the second and third stages, the concentration of oxygen dissolved in the medium pO2, is controlled; rape oil is added in a dose of 20 g/l with increasing the concentration of oxygen dissolved in the medium pO2 to 10%, the aeration is increased to 50-55% to the saturation level.

EFFECT: invention enables increasing the α-ketoglutaric acid to 102,5 g/l.
