FIELD: biotechnology.
SUBSTANCE: said DNA aptamer binds to the bacteria Escherichia coli O157:H7 and is represented as a single-stranded DNA O157Flank with 81 nucleotides in length and molecular weight of 27 kDa. The DNA aptamer has high binding specificity relating to bacteria Escherichia coli O157:H7 and has primary sequence CATACGTTCGACTGCTACTCTGAGTCTGACCTTACAGTCAGTCAATCATGGGACAT ACGAAATGTGAGATGTACAGACTAG. The sequence of the DNA aptamer is selected as a result of positive selection rounds against bacteria Escherichia coli O157:H7 and negative selection against bacteria of other serogroups of Escherichia coli and closely related microorganisms using tRNA as competitor molecule for binding. The DNA aptamer is able to detect specifically the bacteria Escherichia coli O157:H7 in immunoaptamer PCR reaction at a concentration of 103 bacterial cells per 1 ml of solution.
EFFECT: invention enables to detect with high efficiency the said bacteria.
