FIELD: biotechnology.

SUBSTANCE: method involves the preparation of the oligonucleotide combinatorial library of 1015 molecules for selection and breeding oligonucleotide library to a concentration of 1015 molecules per 1 ml denatured oligonucleotides at 90°C for 10 minutes and annealed oligonucleotides at 37°C. Binding of glutathione-S-transferase or streptavidin at 1 mg prepared from the original pool of oligonucleotide library in a buffer containing 20 mM Tris-HCl pH 7.4, 50 mM NaCl and 5 mM EDTA, and separation of the resulting mixture of nucleic acid/target-protein by gel filtration chromatography into fractions.

EFFECT: invention allows to eliminate from oligonucleotide pool maximum possible number of affinity to the molecular predetermined target, and repetition of rounds of such selection warrants in final pool of molecules oligonucleotides having significant for further use affinity for the target, the invention may be applied to obtain vyskoaffine aptamers useful in biomedical field.
