FIELD: medicine.
SUBSTANCE: invention refers to biotechnology, namely to a method for the selective inhibition of PCR amplification of DNA fragments. A reaction mixture is added with a blocking oligonucleotide in the concentration at least twice as much as the PCR primer concentration. The 3′-terminal sequence of the blocking oligonucleotide is used for annealing on the 3′-region of the DNA fragment, the amplification of which is expected to be suppressed, and has a length providing its effective hybridisation on the 3′-region of the DNA fragment at a PCR primer annealing temperature. The 5′-terminal sequence of the blocking oligonucleotide is used as a matrix for filling-in a new 3′-terminus for the above DNA fragment, wherein the new 3′-terminal sequence has no complementarity to the prospective matrixes and eliminates the further involvement of this fragment in the amplification as the PCR primer.
EFFECT: presented invention enables high-effective selective amplification inhibition.
