SUBSTANCE: disclosed is a method and a composition for obtaining a cell characteristic, as well as a kit for producing affinity-oligonucleotide conjugates. Method comprises reaction in drop, wherein the reaction involves attaching a bar-encoded drop polynucleotide to an oligonucleotide portion of the affinity part-oligonucleotide conjugate, where the affine part of the conjugate binds to the individual target cell expressed by the antigen, and the oligonucleotide portion of the conjugate contains a sequence of antigen identification, sequencing the oligonucleotide portion of the conjugate to obtain information on the sequence and determining the cell characteristic in the drop. Composition contains multiple drops, where the individual drop contains a cell, a bar-coded drool polynucleotide and an affinity-oligonucleotide conjugate. Kit comprises a first container comprising a first antigen identification sequence (AID) and a first affinity molecule (AMB) first bar code sequence by a first oligonucleotide, a second container containing a second antigen identification sequence (AID) and a second bar-code sequence of the affinity molecule (AMB) with a second oligonucleotide, as well as reagent containers. Note here that second sequences of AID and AMB differ from first sequences of AID and AMB.
EFFECT: invention provides the possibility of analyzing the antigen of a single cell without the need for cell sorting.
