Characterised methods include introduction of modification into gene spnK or silencing gene spnK with preservation of production of spinosin J and L to eliminate activity of 3'-O-methyltransferase with obtaining genetically modified host cell Saccharopolyspora spinosa, producing spinetoram precursor. Said modification is point mutation, which is located in base pair position, selected from the group, consisting of positions 528, 589, 602, 668, 721, 794, 862, 895, 908, 937 and 1131.

EFFECT: inventions make it possible to produce spinosin with high efficiency.
