SUBSTANCE: fused polypeptide is produced coded by a nucleic acid and containing the following elements in the 5'→ 3' direction, i.e. i) the nucleic acid encoding the polypeptide, ii) the nucleic acid encoding amino acid sequence SEQ ID NO: 01, and the nucleic acid encoding a Strep-tag. Then, the fused polypeptide produced at the previous step is incubated with target-polypeptide, having sequence SEQ ID NO: 02, which is covalently conjugated with poly(ethyleneglycol) fragment at a lysine C-end residue and with a trypsin mutant form D189K, K60E, Na143H, E151H. Then, it is extracted to get the polypeptide conjugated with one poly(ethyleneglycol) fragment.
