FIELD: biology.

SUBSTANCE: presented group of inventions relates to a method for preparing concentrated Brucella cell culture from Brucella abortus 19 strain to prepare brucellous antigens, a method for preparing concentrated Brucella cell culture from Brucella abortus 19 strain to prepare brucellosis diagnostic serum, a method of producing a single brucellar antigen in agglutination test (AT), complement-fixation test (CFT) and prolonged complement fixation test (PCFT), a method of producing brucellar antigen for rose bengal test (RBT), a method of producing brucellar antigen for milk ring test (MRT), a method of producing brucellosis diagnostic serum, a test system for diagnosing brucellosis in animals in AT, CFT and PCFT, a test system for diagnosing brucellosis in animals in RBT and a test system for diagnosing brucellosis in animals in MRT. Described method for preparing concentrated Brucella cell culture from Brucella abortus 19 strain to prepare brucellous antigens used in diagnostic test systems involves obtaining a bacterial cell inoculum and cultivation thereof in deep conditions on a liquid nutrient medium with controlled level of concentration of dissolved oxygen in the culture medium during the whole process of cultivation, inactivation of grown bacterial cells by heating with subsequent concentration. Herewith the process of bacterial cells cultivation is carried out at the temperature of 36-38 °C, in the process of cultivation at the stage of accumulation of bacterial cells from the concentration of 1.5-2.5 billion m.c./cm3 to the concentration of 10 billion m.c./cm3 the concentration of dissolved oxygen in the culture medium is maintained within the range of 20-25 mg/l, and over 10 billion m.c./cm3 and till the bacterial cells reach the stationary growth phase the concentration of dissolved oxygen in the culture medium is provided within the range of 40-45 mg/l, and the concentration of grown bacterial cells is carried out by deposition with a flocculant represented by sodium salt of carboxymethyl cellulose (CMC), which is added to the culture medium in the amount of 0.1-0.15 g as per the dry substance per 1 litre of the culture medium, after which the supernatant is drained to obtain a concentrate of inactivated bacterial cells for further preparation brucellar antigen in AT, CFT, PCFT, RBT antigen; MRT antigen.

EFFECT: invention can be used for diagnostics of brucellosis in animals.
