FIELD: chemistry.
SUBSTANCE: invention relates to a method for efficient production of a polypeptide fragment suitable for binding by NCL method. Method involves a step for reacting polypeptide consisting of a first polypeptide fragment, having at N-terminal a cysteine amino acid residue, and a second polypeptide fragment bonded through an intervening sequence -Cys-W-(His)n-Z-Met- with CNBr to obtain a first polypeptide fragment, having at N-terminal a cysteine amino acid residue, and a third polypeptide fragment, a step for further reaction with a third polypeptide fragment with a compound of formula (I), and a compound of formula (II), to obtain a second polypeptide fragment, having a modified C-terminal.
EFFECT: method is effective for producing multiple peptide fragments; method also enables, compared to traditional method, to perform site-specific glycosylation.
