FIELD: medicine.

SUBSTANCE: cold anaesthesia for the experimental animal is established by quantitative cytochemical evaluation of chromaffin reaction in rat erythrocytes during general body cooling. The method includes cooling of rats, blood taking from the pulmonary vein after phlebotomy in the left lung gate. A blood smear is prepared on a slide, first, stained with 5% alcoholic solution red alizarin C, then 5% solution of potassium bichromate. The number of variously coloured cells in a group of 300 cells and more is counted under the microscope. Then the average cytochemical coefficient ACC is calculated by the formula:

where 1 point - red blood cells not stained; 2 points - granular staining of erythrocyte periphery, cell central part not stained; 3 points - RBC are entirely granularly stained. And at ACC less then 2.4 effective cold anaesthesia is established.

EFFECT: method provides objectification of cold anesthesia assessment for animals, that allows vivisection without application of drug-containing pharmacological agents.
