FIELD: biotechnology.

SUBSTANCE: method for obtaining a chimeric blastocyst is described. Initially, an ultrashort laser pulse is used to fuse two blastomeres of an embryo by irradiating the natural contact site, then a puncture 10-12 microns in diameter is made in the shiny embryo envelope performed in the region located as far as possible from the blastomer by several diode laser pulses, an infrared laser is used to place the pre-modified embryonic stem cells into the puncture.

EFFECT: possibility of rapid and technological introduction of stem cells with a modified genome into a mouse embryo at different stages of development and with different ploidie of blastomeres in a single step, which ensures production of immediately pure lines with higher efficacy, without loss of time and costs for subsequent crossing of chimeras for pure lines excretion, increasing the efficiency of embryos reconstruction.
