SUBSTANCE: invention relates to a method for growing a weakened mutant strain of Salmonella typhi carrying a mutation in the galE gene leading to loss of function and containing at least one copy of a recombinant DNA molecule containing a eukaryotic expression cassette. The invention includes the step of cultivating the strain in a buffer medium containing peptone, at about a neutral initial pH value on a fermentation scale in which glucose is not added to the medium during the fermentation process, and the initial amount of glucose is consumed until reaching a stationary phase where the initial pH is from 5 To less than 9, and wherein the volume of the medium is at least about 10 l.
EFFECT: invention can be used for the industrial large-scale production of DNA vaccines based on attenuated strains of Salmonella.
