SUBSTANCE: described is method of obtaining recombinant purified structural proteins-antigens for production of vaccine against hepatitis B. Cells of yeasts Hansenula polymorpha are transformed with recombinant plasmids, then are cultivated in medium, containing peptone, yeast extract, with forced aeration, biomass control, portioned addition of pure methanol in amount 0.5-1% (volume/volume) relative to fermentation volume into fermentor. Cells are disrupted by extrusion method. Material is purified by diafiltration, precipitation at reduced pH, ion-exchange chromatography, gradient centrifugation, gel-filtration. Together with pH-HBs used are plasmids: pSL-SlS2D2ml33, pSL-HBc-530 or pSL-HBc531-PreSl-3x-PreS2. Combined application of all elements, related to structural proteins of hepatitis B virus, is essential. Invention makes it possible to obtain vaccine, capable of inducing wide range of anti-viral antibodies (anti- preS1, anti- preS2, anti-HBs and anti-HBc), related to different genotypes (serotypes, subtypes) of hepatitis B virus, which makes it possible to apply vaccine in any regions of the world.
EFFECT: due to application of components, specific for preS regions and HBcAg, vaccine can also have therapeutic application.
