FIELD: medicine.
SUBSTANCE: sampling and step-by-step research are performed. During research, distilled water is added to the selected volume of the soil sample. The slurry is stirred and settled for an experimentally set time of 20 minutes or centrifuged for 3 minutes at 1500 rpm. The supernatant is poured into centrifuge tubes and centrifuged for 3 minutes at 1500 rpm. The supernatant is removed from the tubes. The resulting precipitate is examined by an adapted method of immunomagnetic separation followed by immunofluorescence labelling. At that, the number of studies is calculated by the amount of precipitation obtained. The 25 ml of supernatant is placed in 3 tubes, after centrifugation of which 3 sediment samples are formed, calculated as 1 precipitate per 1 study. If the total volume of precipitation in test tubes is not more than 1.0 ml, they are combined into one study, after which the lamblia cysts and the cryptosporidia oocysts are determined by an adapted method of immunomagnetic separation followed by immunofluorescence labelling. The result is taken into account on the basis that lamblia cysts are sparkling and fluorescent apple-green objects, from rounded to oval, 8 to 14 microns in length, 7 to 10 microns in width, with brightly highlighted edges. Cryptosporidia oocysts are sparkling and fluorescent apple-green objects from oval to spherical, 3 to 5 microns in diameter, with brightly highlighted edges.
EFFECT: increased effectiveness lamblia and cryptosporidia oocysts determination in the soil for timely prevention, detection of parasitic contamination.
