FIELD: pharmacology.
SUBSTANCE: method comprises the following steps: gelatine methacrylation by dissolution of a sample of dry gelatine in K-phosphate buffer with stirring on a water bath, incubation of a mixture of gelatine and MA with stirring, K-phosphate buffer application to increase the mixture volume by 1.9-2.1 times, removal of unreacted MA and additional by-products by dialysis against distilled water and subsequent freezing of the resulting solution and lyophilization; preparation of an aqueous solution of silk fibroin by fibroin dissolution in a mixture of CaCl2:C2H5OH:H2O during heating and subsequent dialysis against water, bringing the resulting solution to a concentration of 20-45 mg/ml with water; preparation of a photocuring mixture by dissolution of lyophilized methacrylated gelatine in an aqueous fibroin solution while stirring on a water bath, Irgacure 2959 photoinitiator introduction into the resulting mixture; mixture photocuring by irradiating with UV light (240-300 nm) to obtain hydrogel; obtaining of a fibroin film from hydrogel by providing transition of fibroin molecules from α-helixes and statistical coil in β-layers by hydrogel drying at a room temperature.
EFFECT: adhesion and proliferation of cells, thus providing a possibility of their application in regenerative medicine and bioengineering.
