FIELD: biotechnology.
SUBSTANCE: method of cryopreservation of cyanobacterial cells involves the preparation of a specific nutrient medium for the cultivation of a specific phototropic microorganism, growing it on this nutrient medium. For the cultivation of cyanobacterial cells – Spirulina subsalsa, the Zarrook nutrient environment is used, on which cyanobacterial cells are expanded for 25 days, followed by suspending the grown cells to a biomass concentration of 0.6±0.12 g with 100 % isopropyl alcohol. Freezing is carried out in the presence of 10 % sterile glucose solution. Resulting suspension in a volume of 0.5–0.7 cm3 stand for 25 minutes in the dark with constant stirring at a speed of 20 rpm and placed in cryovials. Cryovials with cyanobacterial cells are placed in pre-chilled to 4 °C polymer container for freezing. Freezing process is carried out at a cooling rate of -1 °C per minute until reaching -80 °C and hold for five hours.
EFFECT: invention makes it possible to increase the survival of cyanobacteria during their freezing-thawing.
