METHOD FOR DETERMINING THE ABILITY OF MYCOBACTERIA OF TUBERCULOSIS TO GROWTH IN ALVEOLAR MACROPHAGES OF PATIENTS AFTER THE COURSE OF ANTITUBERCULOSIS THERAPY Russian patent published in 2018 - IPC C12N5/786 C12Q1/02 C12R1/32

Abstract RU 2652882 C1

FIELD: biotechnologies.

SUBSTANCE: invention relates to biotechnology. Method for determining the ability of tuberculosis mycobacteria to growth in alveolar macrophages of patients after antituberculous therapy is offered. Method involves taking samples of lung tissue from the surgical material, obtaining ex vivo alveolar macrophage cultures from it, fixing the cells on the coverslips 12–20 hours and 5–8 days after the beggining of the culture cultivation, and staining the samples using the Ziehl-Nielsen method. Thereafter, the proportion of alveolar macrophages containing colonies of growing mycobacteria is evaluated and the indicator obtained 12–20 hours after the beginning of the cultivation is compared with the index obtained 5–8 days after the beginning of cultivation. When the proportion of alveolar macrophages containing colonies of multiplying mycobacteria increases from the total number of alveolar macrophages containing mycobacteria, the conclusion about the high ability of mycobacteria to activate reproduction in the alveolar macrophages of the patient is made by 10 % or more during cultivation. With an increase in this indicator less than 10 %, a conclusion about the average ability of mycobacteria to activate reproduction in alveolar macrophages of the patient is made. In the absence of changes in this indicator during the cultivation process or when it decreases, a conclusion is made about the low ability of mycobacteria to activate reproduction.

EFFECT: invention provides higher diagnostic accuracy of tuberculosis.
