SUBSTANCE: invention relates to medicine, namely to plastic surgery, combustiology and can be used in performing skin autoplasty in severely burned patients. Method comprises intensive general treatment, as well as local treatment in the form of necrectomy, wound preparation, autotransplantation with perforation according to MEEK technology and the use of the culture of dermal auto-fibroblasts. In the first 1–3 days after the trauma, a dermal tissue of 0.25–0.35 mm in thickness, 5 cm2 in area, is taken to isolate and cultivate autologous dermal fibroblasts up to the 4th passage. Obtained fibroblasts in a final concentration of 2.0 million cells in 1 ml of physiological saline during the operation before autoplasty irrigate the wound, the dermal surface of the perforation pieces of the skin and the tissue that is included in the MEEK-perforation set on the side of the attached skin. Irrigation is carried out at the rate of not less than 40 thousand cells per 1 cm2 of wound surface and perforated autograft.

EFFECT: method allows to create conditions for rapid engraftment and epithelization of perforated autografts, excludes the risk of transmission of bloodborne infections, allows to reduce the area of donor sites and thereby solve the problem of the scarcity of donor sites in severely burned patients.
