METHOD OF STORAGE OF CELL CULTURES IN SUSPENSION Russian patent published in 2018 - IPC A01N1/02 C12M3/00

Abstract RU 2660075 C1

FIELD: biotechnology.

SUBSTANCE: invention relates to biology and medicine and can be used in the storage of cell cultures. For cryoconservation, use a container with adjustable volume and the possibility of its sealing, at the same time, withdraw the atmospheric gas from the inner volume of the container, and perform the subsequent injection of the volume of the suspension cell culture. Then inject an inert gas into the container volume while increasing the volume of the container to a value providing a selected ratio between the volume of culture and the volume of inert gas placed in the container. Adjustment of the volume of the container is carried out with a rod equipped with a piston. Culture with the gas is mixed in the selected volume of the container at a temperature of +4 to +25 °C for 10 minutes to 2 hours. Next, transfer the cellular suspension saturated with gas to a more compact container with the possibility of sealing, or leave the suspension in a container with a controlled volume. Container is then identified and placed in a freezer with a temperature of -80 °C to -130 °C.

EFFECT: method of cryopreservation of a suspension cell culture ensures the quality and viability of cell cultures while reducing the cost of cells cryopreservation.
