FIELD: chemistry.
SUBSTANCE: invention relates to cell biology and biotechnology, in particular to a method for producing cells expressing a T-cell receptor (TCR). Method comprises bringing cells capable of differentiation into cells of the T cell line into contact with stromal cells and a peptide antigen for differentiation into DN TCRαβ+ thymocytes. Cells capable of differentiation into cells of the T cell line comprise a nucleic acid encoding the TCRα chain from the original TCR, which is specific for the peptide antigen, and the stromal cells comprise a nucleic acid encoding Delta-like-1 or Delta-like-4, and a nucleic acid encoding MHC. Further, a nucleic acid encoding TCRβ chains from TCRαβ+ thymocytes is introduced into the cells obtained earlier, comprising a nucleic acid encoding the TCRα chain and capable of expressing TCR on the cell surface.
EFFECT: invention makes it possible to obtain cells expressing TCR.
