FIELD: organic chemistry.
SUBSTANCE: invention relates to biochemistry, in particular to a genetically modified rodent expressing a humanized T-cell receptor (TCR) and comprising in the genome of its germline: unrearranged TCRα variable gene locus sequence, and/or an unrearranged TCRβ variable locus sequence, to a method for producing thereof, as well as to isolated embryos and tissues. Also disclosed is a method for producing a human T-cell receptor to an antigen of interest, as well as a method for producing a T cell with specificity against an antigen of interest by using said rodent. Invention also relates to a nucleic acid encoding a humanized TCRα polypeptide, as well as to a nucleic acid encoding a humanized TCRβ polypeptide.
EFFECT: invention allows to efficiently obtain a humanized T-cell receptor.
