FIELD: biotechnology.
SUBSTANCE: invention relates to the field of medical biotechnology, namely the method for obtaining and attesting a standard sample of the glycoprotein E of tick-borne encephalitis virus. For this, the tick-borne encephalitis virus is reproduced in a culture of eukaryotic cells, and then inactivated with formaldehyde. Next, the virus is purified and concentrated by precipitation, filtration, tangential ultrafiltration, gel chromatography and repeated ultrafiltration. Attestation of the obtained standard sample is carried out by determining the concentration of the total protein by the spectrophotometric method and the relative concentration of the glycoprotein E of tick-borne encephalitis virus by the method of polyacrylamide gel electrophoresis. Calculation of the absolute concentration of glycoprotein E in a standard sample is made taking into account the concentration of the total protein and the relative concentration of glycoprotein E in the sample. Authenticity of the standard sample is confirmed by the immunoblot method.
EFFECT: invention provides the determination of the quantitative content of glycoprotein E in the sample and the control of the specific activity of the tick-borne encephalitis virus in the tick-borne encephalitis vaccine.
