FIELD: medicine; biotechnology.
SUBSTANCE: invention relates to medical biotechnology, namely the production and control of tick-borne encephalitis virus vaccine (TBE). For this purpose, the cell cultures are infected with strain 205 of the 2nd passage tick-borne encephalitis culture virus with the subsequent reproduction of the virus on the Vero cell line; to increase the reproduction of the virus, glutamine, fetal calf serum is added. Cultivation is carried out in roller bottles or cell factories, after which the viral suspension is inactivated with formaldehyde. Next, the suspension is concentrated on hollow fiber or membrane modules with a cut-off threshold of 100–300 kDa, followed by chromatographic purification using a polymeric sorbent and sorbing on an adjuvant. DNA concentration of producer cells in the vaccine does not exceed 10 ng/dose, the concentration of cattle blood plasma proteins does not exceed 50 ng/dose.
EFFECT: invention provides a vaccine that contains the minimum amount of heterogeneous proteins and has increased safety, efficacy and stability.
