FIELD: medicine.
SUBSTANCE: group of inventions refers to immunology, medicine, pharmacology, and can be used for production of inactivated influenza vaccines. Proposed method for producing the influenza virus antigen or antigens includes production of chicken embryos (CE), their disinfection, pre-incubation, infection with a working solution of the seed virus, incubation of the infected CE, cooling after incubation, collecting a virus-containing allantoic liquid, disaggregating the recovered viruses by adding a NaCl solution to a final concentration of 0.23–0.30 M, inactivation by adding β-propiolactone to final concentration of 0.1 % and purification of virus-containing allantoic liquid by microfiltration using cascade of three filters with pore diameter of 10 mcm, 1 mcm, 0.6 mcm and ultrafiltration with cutoff limit of 300 kDa, then concentrated clarified virus-containing allantoic fluid is not more than 20 times, concentrate is purified by way of double ultracentrifugation in sucrose density gradient, after which inactivated viral concentrate is split using detergent n-octyl-β-D-glucopyranoside (octylglucoside) or tetradecyltrimethylammonium bromide (TDTAB), removal of undamaged virions and complexes of ribonucleoproteids with membrane protein by centrifugation, micro- and ultrafiltration, chromatographic purification and sterilizing filtration of the obtained antigen or antigens, and the antigen or antigen is stabilized using a Triton X-100 detergent in concentration of 100–200 mcg/ml. What is also presented is an influenza vaccine containing an antigen or antigens prepared by the above method.
EFFECT: development of industrial method for obtaining antigen and / or antigens of high-purity virus for production of inactivated vaccine, which can be used for mass vaccination of population.
