FIELD: biochemistry.
SUBSTANCE: invention relates to biochemistry, particularly to a modified IgG4 Fc-fragment, having a low risk of inducing chain exchange and formation of a monomer in vivo, as well as a nucleic acid which encodes it. Also disclosed is a vector, a host cell containing said nucleic acid, as well as a method of producing an Fc-IgG4 fragment using said cell. Invention also relates to a conjugate of a medicinal agent containing said Fc-fragment of IgG4 conjugated with a drug by a linker.
EFFECT: invention enables effective minimization of the effector function of IgG4 Fc and the exchange of chains with IgG in vivo and can maintain in vivo activity of the drug conjugate and improve its prolonged release in vivo.
