FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology. To solve the task, optimal fragments of the genome of each microorganism, as well as human DNA, are determined from the position of specificity: obtaining a set of oligonucleotide primers and probes for these fragments for LAMP, designed so that to be aimed at six specific regions of the target gene, which are designated as F3c, F2c and F1c by 3'-terminal and B1c, B2c and B3c by 5'-terminal, first external primer F3 consists of F3 region, which is complementary to F3c fragment of target gene, second external primer B3 consists of B3 region, which is complementary to B3c section of target gene, third primer - internal FIP contains F2 fragment by 3'-terminal. which is complementary to F2c, as well as the same sequence, as F1c target DNA region by 5'-terminal, fourth BIP primer is also internal and consists of region B2 by 3'-terminal and same sequence as B1c region by 5'-terminal, and probes, for quantitative detection of coccal microflora using the LAMP method, based on an automatic cycle of DNA synthesis of the chain with mixing using DNA polymerases with high bias activity and four or six specially created primers; method is developed for calculating dysbiotic states by the LAMP threshold cycle.
EFFECT: creation of a molecular-genetic panel for accelerated high-specific detection of dysbiotic disorders of skin and mucous membranes by isothermal PCR using common PCR equipment.
