SUBSTANCE: invention relates to biotechnology and specifically to a method and apparatus for determining an analyte in a sample through a closed flow cell. Device for selective enrichment of a population of X-bearing cells or Y-chromosome carriers in a liquid sample containing spermatozoa of non-human mammals includes a closed flow cell, comprising: sample inlet opening, specimen outlet opening, current path defined between inlet opening and sample outlet opening, wherein the current path passes through the examination area and the sorting section located after the examination area, where at least part of the liquid flow passing from the uppermost part of the investigation area to the lowest part of the sorting section is limited by the wall of the flow cell to form a continuous closed liquid flow path, without separation or branching; hydrodynamic focusing element located in front of the investigation zone and creating a core-in-shell-type flow along the flow axis between the examination area and the sorting section, wherein the core-in-envelope flow comprises a nucleus of a flow carrying spermatozoa of mammals not belonging to the human genus, and hydrodynamic focusing element is configured to maintain a maximum dissipation rate of local energy (EDR) experienced by sperm cells below a threshold value; concave ellipsoidal collecting element having an apex, an optical axis, a first focusing point located in the current path investigation zone, and a second focusing point; lighting means comprising a plurality of electromagnetic energy beams converging nonaxially in the examination area, wherein each of said beam combination is perpendicular to the flow axis; and a direct destructive radiation source configured to deliver electromagnetic energy to the core of the flow when the flow core passes through the sorting section.
EFFECT: technical result is higher sensitivity and efficiency of the method and device for identification and/or separation of cells.
