METHOD FOR DETERMINING THE RELATIVE AMOUNT OF ETHOTICALLY TRANSFORMED PHAGOCYTES Russian patent published in 2020 - IPC G01N33/48 G01N33/49

Abstract RU 2712179 C1

FIELD: medicine.

SUBSTANCE: invention refers to medicine, namely to a method for determining a relative amount of ethotically transformed phagocytes. Method consists in carrying by end of smearing surface of plate on working surface of working glass, on which smear is applied, wherein blood smear is made uniform in thickness of one layer of erythrocytes with erythrocyte density in it 4–15 thousand per mm2, wherein to determine the amount of ethotically strongly transformed phagocytes in preparing the smear in blood, albumin is added in concentration of 1–5 %, to determine the amount of all ethotically transformed phagocytes, the blood sample for the smear is mixed with isotonic salt solution, and blood content in the mixture ranges from 25 % to 75 %, and to determine total amount of medium and strongly ethotically transformed phagocytes whole blood is analyzed; smears are dried for 5–30 s; working glasses for preliminarily calibrating smears, wherein control blood smears are made of at least 10 healthy donors, total amount of ethotically medium and strongly transformed phagocytes in each smear is determined, and when determining their values within 1–11 % of not less than 95 % of results and their arithmetic mean value within 4–6 %, such glasses are used for preparing smears; and to determine the relative amount of ethotically transformed phagocytes in the test smear, the amount of native phagocytes is determined, and the ratio of the corresponding ethotically transformed phagocytes to the sum of these ethotically transformed phagocytes and native phagocytes is determined by the relative amount of the corresponding ethotically transformed phagocytes.

EFFECT: method described above enables higher accuracy of determining the number of ethotically transformed phagocytes.
