SUBSTANCE: group of inventions relates to the field of analytical chemistry. Disclosed is a method of detecting the presence or absence of a target molecule in a sample, comprising bringing the sample into contact with a molecule comprising a cleavable linker, wherein said cleavable linker is specifically cleaved in the presence of said target molecule; loading said sample into a device comprising a nanopore; creating a configuration of the device with the possibility of passing a frame selected from nucleic acids, peptide nucleic acid (PNA), dendrimers, linearized proteins or peptides, through said nanopore, a cleavable linker containing a first domain which binds to said framework, and a second domain which binds to the molecular weight, thereby forming a complex, and wherein the device comprises a current sensor; and determination using sensor, whether the splitted linker was split by detecting an individual current signature during translocation of said complex through the nanopore. Also disclosed are alternative methods for detecting a target molecule, a method for quantitative determination of a target molecule and a kit for realizing said methods.
EFFECT: group of inventions enables to carry out a single-molecular method of detecting the presence of any active enzyme which splits a specifically cleavable linker associated therewith, as well as facilitating detection of enzyme activity, since the added mass in the form of molecular load creates a clear difference of signatures between the complex, the frame and the load.
