FIELD: medicine; pharmaceuticals.
SUBSTANCE: invention refers to pharmaceutical industry, namely to a sterile transparent viscous-flow water-salt solution for making biomedical cell products or three-dimensional fabric engineering structures. Sterile transparent viscous fluid water-salt solution for making biomedical cell products or three-dimensional fabric engineering structures containing acid, purified collagen retains the ability to form fibrils and stable hydrogels in physiological conditions. Method of producing a collagen solution involves successive stages: 1) extraction of collagen-containing tissue under certain conditions; 2) multiple purification of acidic water-salt extract from potentially immunogenic macromolecules; 3) purification of acidic water-salt extract from remaining impurities of potentially immunogenic macromolecules; 4) purification from pyrogenic impurities; 5) purification from impurities of common and spore-forming microorganisms, as well as supramolecular aggregates of collagen; 6) bringing collagen solution to required concentration of collagen protein. Method of producing a collagen solution involves successive stages: 1) extraction of collagen-containing tissue in diluted organic acid solution; 2) multiple purification of acidic water-salt extract from potentially immunogenic macromolecules; 3) purification of acidic water-salt extract from remaining impurities of potentially immunogenic macromolecules; 4) purification from pyrogenic impurities; 5) purification from impurities of common and spore-forming microorganisms, as well as supramolecular aggregates of collagen; 6) bringing collagen solution to required concentration of collagen protein in it. Disclosed is use of collagen solution as initial material for formation of biopolymer matrix with inclusion of living cells; use of collagen solution for production of bioink for 3D printing with resolution of parts up to 0.3 mm and bioprinting of complex structures with inclusion of living cells; use of a collagen solution to produce stable transparent polymer hydrogels which support growth and differentiation of cells in a three-dimensional culture; use of collagen solution for production of stable transparent polymer hydrogels used as material for creation of transparent collagen membranes suitable for implantation.
EFFECT: disclosed group of solutions is efficient, safe, enables to create structures using 3D without using chemical and/or photochemical cross-linking.
