SUBSTANCE: group of inventions relates to a mechanism for intracellular and intramembranous delivery of therapeutic polypeptides. Extracellular vesicle (EV) for intracellular polypeptide delivery contains at least one polypeptide structure containing: (i) at least one polypeptide of interest (Pol); (ii) at least one exosomal polypeptide and (iii) a polypeptide-based release system, which releases said at least one Pol from at least one exosomal polypeptide in endogenous activation, wherein said at least one Pol is bound to the exosomal polypeptide through an endogenously activated release system. Also disclosed are a method for producing an EV, a polypeptide construct for producing an EV, a polynucleotide structure for producing EV, cell for producing EV, an in vitro method for intracellular delivery of at least one polypeptide of interest, a pharmaceutical composition, use of EV in medicine.
