SUBSTANCE: invention relates to the field of biochemistry and genetic engineering, in particular to a constructed single guiding nucleic acid. This nucleic acid targets a nucleic acid and is capable of complexing with the Cas9 CRISPR II type protein. Present nucleic acid comprises a spacer sequence, a duplex, a linker sequence, and a tracr sequence, and is characterized in that it does not contain the region of the first stem. Invention also relates to a composition and to a kit that contain said nucleic acid.

EFFECT: present invention makes it possible to obtain a constructed single guiding nucleic acid targeting a nucleic acid and capable of complexing with the Cas9 CRISPR II type protein.
