SUBSTANCE: disclosed is a method for synthesis of a RNA molecule of a given sequence, comprising synthesis and use of RNA molecules of a reaction mixture, where the reaction mixture is optimized for a given sequence such that it includes four ribonucleoside triphosphate (NTP) GTP, ATP, CTP and UTP so that the fraction of each of them corresponds to the fraction of the corresponding nucleotide in the RNA molecule. Described is an optimized mixture, its use for synthesis of an RNA molecule of a given sequence and a kit containing an optimized mixture, a buffer, a DNA matrix and RNA polymerase.
EFFECT: this group of invention provides more effective RNA transcription in vitro.
