FIELD: cell biology.
SUBSTANCE: method of producing T-cells and a method for producing a therapeutic agent based on human T-cells, providing for culturing a population of peripheral blood mononuclear cells (PBMC), which contains T-cells and antigen-presenting cells (APC), for 20 hours to 24 hours in a medium containing exogenous interleukin-2 (IL-2), CD3 antibody or CD3-binding fragment thereof and CD28 antibody or its CD28-binding fragment, B7-1 or its CD28-binding fragment, or B7-2 or its CD28-binding fragment; and transducing a PBMC population by a lentiviral vector comprising a polynucleotide encoding a chimeric antigenic receptor (CAR) to a B-cell maturation antigen (BCMA).
EFFECT: this group of inventions can be used in production of adoptive therapy means.
