FIELD: biotechnology.
SUBSTANCE: present set of inventions relates to analysis of nucleic acids. Disclosed are methods for sequencing templates on the basis of nucleic acid which comprise providing array of sites comprising a mixture of different templates and extending primers comprising different reversible blocking groups and being hybridized to different templates, which leads to producing different primer extension products at each site as well as detecting different primer extension products and removing different reversible blocking groups from primer extension products with said actions being repeated with the aim to determine sequence of different nucleotide analogs attached to each of different extension products at each site orthogonally. Besides, disclosed is an array of sites of nucleic acids relating to said primers for use in said methods.
EFFECT: use of selective and orthogonal deblocking of templates at the same site of the array enables increasing in collection of information from the array by at least two times.
