FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology and can be used to check the quality of samples prepared for high-throughput DNA sequencing. To determine efficiency of NGS-library enrichment with exon sections for human exome sequencing qPCR with human DNA of the same NGS-library is performed before and after enrichment procedure. Each PCR reaction is carried out with at least 50 pg DNA of the NGS library. Efficiency of the qPCR reaction and the threshold cycles of the fluorescent signal of the NGS-library samples before and after the enrichment procedure are determined. Enrichment efficiency of the NGS library is then calculated using the given formula.
EFFECT: technical result consists in improvement of NGS-library enrichment efficiency evaluation accuracy.
