SUBSTANCE: invention relates to medicine, biology, biotechnology, clinical-laboratory diagnostics, molecular biology, genomics and genetics and is a method for enriching a DNA fragment library for subsequent sequencing by high-throughput second-generation sequencing (NGS) protein-coding sequences using duplex-specific thermally stable nuclease of king crab. Hybridisation involves addition of an additional portion of human DNA pre-enriched with repeating DNA sequences including members of the Alu and Kpn family.

EFFECT: invention increases sequencing efficiency.
