Abstract RU 2750382 C1

FIELD: chemical and pharmaceutical industry; medicine.

SUBSTANCE: present invention relates to the field of chemical and pharmaceutical industry and medicine, namely, to a method for producing an immobilized enzyme preparation based on collagenase, polysaccharides modified with vinyl monomers, and hyaluronic acid. The method includes the dissolution of collagenase in an aqueous solution of low-molecular-weight hyaluronic acid 300 kDa or medium-molecular-weight hyaluronic acid 500 kDa or high-molecular-weight hyaluronic acid 800 kDa in a ratio of 10 mg of collagenase per 2 ml of an aqueous solution of low-molecular-weight hyaluronic acid 300 kDa or medium-molecular-weight hyaluronic acid 500 kDa or high-molecular-weight hyaluronic acid 800 kDa at a concentration of 1.5%. Mixing is carried out until complete dissolving at room temperature. Collagenase is then immobilized by adding to the resulting mixture of a graft copolymer of carboxymethylcellulose 50-100 kDa (CMC) or chitosan 50-100 kDa (CHTS) with N-vinylimidazole (VI) or N,N-dimethylaminoethylmethacrylate (DMAEMA) in an amount of 100 to 290 mg to obtain a liquid preparation or 300 to 500 mg to obtain a gel. Due to the invention, collagenase solutions can be stored at temperatures from 4 to 25℃ for 21 days, and the enzyme becomes more stable in comparison with the native one and it is able to penetrate into the deep layers of the skin. The introduction of modified polysaccharides into the composition of the preparation allows not only to vary the viscosity and consistency of the samples (from the liquid state to the gel with different fluidity), but also promotes the formation of conjugates with an enzyme with a large number of bonds and interactions between the polysaccharide and the enzyme, compared with unmodified carboxymethylcellulose and chitosan. This further increases the stability of the preparation and the concentration of the active substance in the affected area of the skin, and provides a controlled (portion) release of collagenase, maintaining its necessary concentration for a long time.

EFFECT: technical result is an increase in the stability of the preparation.
