SUBSTANCE: invention relates to an in vitro method for determining a modulating impact of an examined compound on epithelium barrier function, which includes stages of a) providing a micro-fluid system containing a set of hollow micro-fluid canals, while the specified system contains at least one canal, which is at least partially filled with hydrogel; b) introducing epithelium cells in the specified micro-fluid canal and providing bringing the specified epithelium cells into contact with the specified hydrogel; c) cultivating the specified epithelium cells, which were introduced in the specified micro-fluid canal, thus allowing the specified cells to form on hydrogel a cell layer with apical and basolateral sides; d) providing the specified epithelium cells in the specified micro-fluid canal with a probe and the specified examined compound, while the specified probe and the specified examined compound are provided independently from apical side, from basolateral side or from both apical and basolateral sides; e) determining in different moments of time a signal provided by the specified probe in the specified micro-fluid canal or in the specified hydrogel, or both in the specified micro-fluid canal and the specified hydrogel, and thus determining the modulating impact of the examined compound on the epithelium barrier function.
EFFECT: invention provides for a possibility of determining, measuring, analyzing, predicting or establishing the negative or positive impact (effect) of the compound on the epithelium barrier function.
