FIELD: biotechnology.
SUBSTANCE: present invention relates to the field of biotechnology, in particular to methods for producing antibodies. The invention is aimed at the optimization of the production of selectively capped and uncapped cysteines in antibodies by manipulating cell growths conditions, including intentional depletion of cysteine and/or cystine in the process of cell cultivation using medium components, duration of batch cultivation or cell density, to achieve effective protein production.
EFFECT: invention allows for obtaining a cysteine-mutant trastuzumab antibody K290C-K334C or K392C-K334C, which can further be conjugated with chemical useful load.
