FIELD: medicine; molecular biology.
SUBSTANCE: invention relates to medicine and molecular biology, namely to methods for molecular genetic detection of SARS-CoV-2 coronavirus RNA, as well as quantitative assessment of viral load in autopsy, biopsy and surgical material during pathoanatomic studies by quantitative polymerase chain reaction in real time (qPCR). A set of primers and probes for the identification of coronavirus cDNA is described. Also described is a method for measuring the viral load of SARS-CoV-2 in tissues of various organs by quantitative polymerase chain reaction in real time, which is based on detecting the number of copies of SARS-CoV-2 cDNA relative to 100 copies of ABL1 cDNA. The invention provides an increase in the reliability of quantitative assessment. Serial dilutions of pGEM®)-T Easy plasmid DNA containing sections of SARS-CoV-2 and ABL1 cDNA can be detected with a high correlation of linear dilutions equal to R2≥0.99.
EFFECT: developed oligonucleotide primers and fluorescently labeled probes can be used to detect and quantify the viral load of SARS-CoV-2 in the tissues of various organs.
