METHOD AND COMPOSITION FOR ANALYSIS OF CELLULAR COMPONENTS Russian patent published in 2021 - IPC C12Q1/6806 C12N15/10

Abstract RU 2761432 C2

FIELD: biotechnology.

SUBSTANCE: proposed is a method for analysis of cellular components (variants). In the first variant, the method includes combining lysis buffer drops and cellular suspension drops, combining lysate drops with reagent drops, wherein the reagent includes a reporter group and enzymes, combining drops including the reporter group with drops of a washing solution, and obtaining the sequence information. In the second and third variants, the method includes applying cells to solid substrates, wherein each solid substrate constitutes a cohesive element (CE), conducting cell lysis forming multiple CEs, compartmentalising the CEs forming first compartments, providing a compartment of a first reporter group and a modification of a nucleic acid, combining the CEs, compartmentalising the CEs forming second compartments, providing a compartment of a second reporter group and a modification of a nucleic acid. In the second variant, the method includes obtaining sequence information from a nucleic acid including the first reporter group, and in the third variant, the method includes analysis of one or more first reporter groups and one or more second reporter groups of substances.

EFFECT: inventions provide a possibility of distinguishing between identical molecules after processing and determining the initial representation of molecules or a population of molecules.
