FIELD: chemistry.
SUBSTANCE: group of inventions relates to methods for isolating specific agents of interest (hereinafter – AOI), such as proteins that are optionally present or produced in a culture. Methods are based on reversible binding of chimera consisting of a cellulose-binding domain (CBD) and an agent of interest (AOI), such as protein, for example heterologous protein (HP), with cellulose nanocrystals (hereinafter – CNC) to create large protein/CNC adducts that are separated from contaminants by filtration through ceramic hollow fiber membrane or polymer hollow fiber membrane. Then, AOI, for example protein, can be isolated from a complex.
EFFECT: inventions provide for a quick and effective three-stage method for purifying/isolating AOI using filtration devices and technologies, allow for processing of very large volumes of solutions with a wide spectrum of proteins and solid particles, providing a very high degree of selectivity, exclude the need for affine chromatography.
