FIELD: cellular engineering.
SUBSTANCE: invention relates to the field of cellular engineering, in particular to a method for creating cell transplants for the treatment of type 1 diabetes mellitus. To implement the specified method, insulin-producing cells and MMSC of adipose tissue are obtained by enzymatic tissue treatment using a solution of 1A collagenase at a concentration of 0.1% with the addition of a Hanks solution and a cultivation medium with the addition of 15 mM of HEPES. After that, MMSC of adipose tissue are planted on substrate in the amount of 10×103 NC (nucleated cells)/cm2, cultivating for the purpose of building up and cleaning from non-target cells for 10-14 days. Islets obtained from the pancreas are centrifuged on a cascade of density gradients using ficoll-urographin to clean islets from pancreatic tissues and non-target cells. Further, islets are disaggregated by fermentation in a 0.25% Trypsin-EDTA solution, followed by centrifugation with a Hanks solution with the addition of 15 mM of HEPES. Then, insulin-producing cells are planted on substrate at a concentration of 10×104 NC/cm2 and cultured for 10-14 days with alternating DMEM media with a glucose content of 1 g/l for 8 days and a glucose content of 4.5 g/l for 2-6 days. After cultivation, insulin-producing cells are mixed with MMSC of adipose tissue in a ratio of 1:1 and encapsulated in sodium alginate of 1% low-viscosity, polymerized with the addition of BaCl2 at a concentration of 2.2%, with the addition of saline – 0.9% sodium chloride.
EFFECT: present invention makes it possible to reduce immunogenic properties of insulin-producing structures, which helps to avoid fibrosis and the graft-versus-host reaction in the treatment of type 1 diabetes mellitus.
