FIELD: biotechnology.
SUBSTANCE: group of inventions relates to recombinant strains of Mycolicibacterium neoaurum, as well as use thereof to produce 22-functionalised steroids. What is presented is a recombinant strain Mycolicibacterium neoaurum VKM Ac-2846D, which transforms sterols into 24-norhol-4-ene-3,22-dione, 20-hydroxymethyl-pregn-4-en-3-one and corresponding 1-dehydroanalogues. Also disclosed is a recombinant strain Mycolicibacterium neoaurum VKM Ac-2847D, which transforms sterols into 24-norhol-4-ene-3,22-dione and 20-hydroxymethyl-pregn-4-en-3-one without admixture of corresponding 1-dehydroanalogues. Said strains are obtained by introducing a marked directed deletion into the steroid gene β-ketoacyl-CoA-thiolase fadA5 in corresponding parental strains. Said strains are used for biotransformation of phytosterol into 24-norchol-4-ene-3,22-dione, 20-hydroxymethyl-pregn-4-en-3-one and their 1-dehydroanalogues.
EFFECT: disclosed are recombinant mycobacterial producer strains and use thereof to produce 22-functionalised steroids.
