FIELD: pharmacology.
SUBSTANCE: invention relates to the field of pharmacology and can be used to block the recurrence of infectious diseases and enhance the effect of vaccines. The proposed composition of immunostimulating substances of muramyl peptide nature, containing NOD2-receptor ligands, includes as substances of muramyl peptide nature a mixture of substances isolated from an aqueous extract of muscles and skin of the grass snake Natrix natrix by preparative separation using thin-layer chromatography, using silica gel as a carrier during separation in a mixture of isopropanol: water = 1:1, with the formation of spots on the chromatogram with values Rf = 0.06 and Rf = 0.57, while the gross formula of the isolated substances, corresponding to the spot on the chromatogram with a value of Rf = 0.06 correspond to C22H33O2N6 or C19H35O5N5 with M=413.26448 Da, as well as C20H37O2N6 or C17H39O5N5 with M=393.2960 Da, and the gross formulas of the isolated substances corresponding to the spot on the chromatogram with values Rf = 0.57 correspond to C22H43O8 or C20H41O7N3, or C18H39O6N6 with M = 435.29441 Da. At the same time, this aqueous extract of the muscles and skin of the grass snake was obtained by boiling the cooked snake carcass in a 0.1 N HEPES buffer pH 7.0-7.2, pre-purified with chloroform, then freeze-dried under vacuum at a temperature from 0 to - 2°C to obtain a powdered preparation, diluted in water and purified using a series of chromatographic cleanses: on DEAE-Sepharose in the same buffer with the selection of a fraction that is not sorbed on this carrier in this solution, using gel filtration on Sephadex G-15 or G-25 with the selection of the contents of the first two peaks at λ = 220 nm from those separated on this carrier and mixing of the selected material, on column C18 Discovery with the selection of the material of the first expressed peak at λ = 220 nm with concentration of contents under vacuum, on a column with NH2-Sepharose with collection of the first peak, visible both at λ = 220 nm and at λ = 280 nm.
EFFECT: increase in adjuvant activity for therapeutic or preventive use of vaccines.
